摘要
目的:观察威灵仙脂质体乳膏对兔膝骨关节炎关节液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量及关节形态学的影响。方法:将32只白兔随机分为正常组8只和造模组24只,造模组采用关节腔内注射木瓜蛋白酶法建立OA模型,成模后随机分为模型组、威灵仙脂质体乳膏组和扶他林乳膏组,每组8只。采取膝关节处涂抹乳膏的方法治疗8周。治疗结束后收集关节液及关节软骨标本,用放免法检测关节液中TNF-α、IL-1β的含量。观察兔膝骨关节组织病理学变化,并用Colombo评分比较各组差异。结果:威灵仙脂质体组关节液中TNF-α、IL-1含量以及关节软骨Colombo病理学评分显著降低(与模型组比,P<0.01),光镜下其软骨结构保存基本完整,表面溃疡糜烂较轻微,软骨细胞破坏不严重,电镜下细胞膜及细胞核基本完整,内质网病变较轻。结论:威灵仙脂质体能抑制骨关节炎兔关节液中TNF-α、IL-1β的产生,对兔膝OA的损伤软骨具有较好的修复作用。
Objective:To observe the effects of Radix Clematidis Liposomes (RCL) on the content of TNF-α and IL-1β in the synovial fluid and the morphological changes in knee joints of osteoarthritis (OA) rabbits. Methods: Thirty-two New Zealand rabbits were randomly divided into normal control, model group, RCL cream group and Volalin cream group, 8 rabbits in each group. The OA models were established by intra-articular injection of papain. After the treatment of 8 weeks, the samples of the synovial fluid and articular cartilage were collected. The levels of TNF-α and IL-1β in synovial fluid were measured by RIA assay. Pathologic changes of the cartilage were observed and graded according to the Colombo histological score. Results:The content of TNF-α, IL-1β and Colombo scores of articular cartilage were significantly de creased in RCL cream group compared to model group (P〈0.01). Under the light microscope, the structure of cartilage remained complete generally, with mild ulcer and anabrosis on the surface and non-severe damage of the cartilage cells; under the electro-microscope, the cell membrane and nucleus were almost intact, and the damage of the endoplasmic reticulum was slight. Conclusion:The RCL can inhibit the production of TNF-α and IL-1β in synovial fluid and improve the recovery of damaged cartilage in knee joints in OA rabbits.
