星形诺卡菌对多巴胺能细胞损伤的体外研究

摘要 目的探讨与帕金森病可能相关的星形诺卡菌导致多巴胺能细胞损伤的可能机制。方法测定星形诺卡菌作用于多巴胺能细胞系SH-SY5Y后细胞存活率的变化、线粒体呼吸链复合物Ⅰ活性及细胞内活性氧、线粒体膜电位的改变,并与MPP+所致的多巴按能细胞损伤进行对比。结果星形诺卡菌作用于多巴胺能细胞系SH-SY5Y,可致细胞存活率显著下降;星形诺卡菌与细胞共孵4~38h,细胞内活性氧减少、膜电位降低、线粒体呼吸链复合物Ⅰ活性显著下降。结论氧化应激及线粒体功能受损是导致多巴胺能细胞损伤的可能机制,最终致细胞变性死亡。 Objective To study the possible mechanism of dopaminergie ceils death induced by Nocardia asteroids which may play a great role in the pathogenesis of Parkinson disease. Methods The cellular survival rate,the activity of mitochondrion respiratory chain compound I , the level of cellular Activated oxygen and membrane potential of mitochondrion were measured in dopaminergic cell line SH-SY5Y after the action of No cardia asteroids, which were be compared with the dopaminergic cells after MPP^+ exposure. Results The cellular survival rate of dopaminergic cell line SH SY5Y was dramatically decreased by the action of the Nocardia asteroids; meanwhile the level of cellular Activated oxygen,membrane potential of mitochondrion and the activ ity of mitochondrion respiratory chain complex I were also markedly descent after co-incubation of Nocardia asteroids and cells for about 4438 hours. Conclusions The mechanism of dopaminergie cells injury induced by Nocardia asteroids may be oxidative stress and the damage of function of mitochondrion, which may lead to do paminergic cells degeneration and apoptosis.
