过氧化物酶体增殖受体γ辅激活因子-1多态性与2型糖尿病相关性研究 被引量:1

摘要 [目的]探讨延边地区朝鲜族、汉族人群中过氧化物酶体增殖受体γ辅激活因子-1(PGC-1)多态性与2型糖尿病的相关性。[方法]采用聚合酶链反应-单链构象多态性(PCR-SSCP),对104例正常人群和113例2型糖尿病患者的PGC-1基因Thr394Thr、Leu410Iso和Leu438Ser多态性基因型进行对照分析。[结果]朝鲜族和汉族正常人群PGC-1基因Thr394Thr多态性的A等位基因频率分别为23.3%和20.7%,两民族间无统计学差异;糖耐量正常组的PGC-1基因Thr394Thr多态性的A等位基因频率为22.1%,2型糖尿病组的A等位基因频率为22.1%,两组间无差异;未检测出Leu410Iso和Leu438Ser多态性。[结论]PGC-1基因Thr394Thr多态性在延边朝鲜族和汉族人群之间无差异;PGC-1基因Thr394Thr与2型糖尿病无相关性。 [Objective] To determine the relationship of peroxisome proliferator-activated receptor-ycoactivator 1 (PGC- 1 ) gene polymorphism with type 2 diabetes mellitus for Korean-Chinese nationality and Han-Chinese nationality in Yanbian area. [Methods] PCR-SSCP was used to test PGC-1 gene Thr394, Leu410Iso and Leu438Ser polymorphism in 104 cases with normal glucose tolerant (NGT), and 113 cases with type 2 diabetes mellitus (T2DM) of Korean-Chinese nationality in Yanbian area. [Results] The A allele frequencies of NGT were 23.3% for Korean-Chinese nationality, and 20.7% for Han-Chinese nationality, and no significant difference was found between two groups. The A allele frequencies of NGT and T2DM were both 22.1% for ordinary people and patients, and no significant was found between the two groups. The Leu410Iso and Leu438Ser polymorphism were not detected in any cases. [Conclusion] There was no difference in frequencies distribution of Thr394Thr polymorphism of PGC-1 gene between Korean-Chinese nationality and Han-Chinese nationality in Yanbian area, and the Thr394Thr polymorphism of PGC-1 gene has no association with T2DM.
