摘要
目的建立同时测定痔痛安搽剂中三种蒽醌类成分大黄素、大黄酚和大黄素甲醚含量的方法,提高制剂的质量控制标准。方法采用HPLC法。色谱柱为Diamonsil C18柱,流动相为甲醇-质量分数为0.1%的磷酸水溶液(体积比75:25),流速1.0mL·min^-1,检测波长254nm,柱温35℃。结果大黄素、大黄酚、大黄素甲醚质量浓度分别在0.64~20.48mg·L^-1(r=0.9995)、0.31~9.84mg·L^-1(r=0.9997)、0.50~16.08mg·L^-1(r=0.9998)内与峰面积呈良好的线性关系;大黄素平均加样回收率为98.8%,大黄酚平均加样回收率为99.6%,大黄素甲醚平均加样回收率为98.5%。结论采用HPLC法可同时测定痔痛安搽剂中三种蒽醌类成分大黄素、大黄酚和大黄素甲醚的含量,且操作方法简便,分离效果好,运行成本低。该法可用于痔痛安搽剂的质量评价。
Objective To develop a method for determination of emodin, ehrysophanol and physeion in Zhitong' an(traditional Chinese medicines) liniment. Methods RP-HPLC method was used with Diamonsil C18 (250 mm×4.6 mm, 5 μm) column. The mobile phase was methanol-0.1% phosphoric acid ( V: V= 75:25), flow rate was 1.0 mL·min^-1. The detection wavelength was set at 254 nm, and column temperature was 35 ℃. Results The calibration curves were linearin the range of 0.64- 20.48 mg·L^-1 (r = 0.999 5)for emodin, 0.31-9.84 mg·L^-1(r =0.999 7) for ehrysophanol and 0.50-16.08 mg·L^-1(r= 0. 999 8) for physeion. The average recoveries of emodin, physeion and ehrysophanol were 98.8 %, 99.6 % and 98.5%, respectively. Conclusions The method is convenient and appropriate for determination of emodin, ehrysophanol and physeion in Zhitong' an liniment.
