摘要
[目的]初步探讨肥胖SD大鼠肝脏组织中细胞因子信号传导抑制因子3(suppressor of cytokine signaling 3,SOCS3)mRNA的含量与高瘦素(leptin)血症的关系。[方法]采用高脂饲料喂养以诱导的肥胖(DIO)SD大鼠模型,应用放射免疫分析法检测血清leptin水平,应用RT-PCR半定量法检测肝脏组织中SOCS3 mRNA的含量,并进行相关分析。[结果]DIO组大鼠存在高leptin血症,其肝脏组织中SOCS3 mRNA的含量0.9392±0.0081明显高于正常饮食组大鼠0.3460±0.0743,两组间差异有显著性意义(P<0.01),而且肝脏组织中SOCS3 mRNA含量与大鼠的肥胖程度及血清leptin水平呈显著正相关,P<0.01。[结论]肝脏组织中SOCS3可能是DIO大鼠的leptin抵抗的机制之一。
[Objective] To investigate the relationship between the expression of liver (suppressor of cytokine signaling 3,SOCS3) mRNA and serum leptin concentration.[Methods] To duplicate the obese rat model by feeding with high-fat diet(diet-induced-obesity rats,DIO group),and utilize the radioimmunity to deteet the serum leptin concentration,semi-quantitative RT-PCR to measure SOCS3 mRNA value in rat liver tisrue and then analyze the relationships between them.[Results] In DIO group,the concentration of serum leptin 0.9392±0.0081 was higher than that in control group with normal diet 0.3460±0.0743,and the expression of SOCS3 mRNA in liver tisscce of DIO group is also higher than that of cowtrol with significant difference(P〈0.01).The expression of SOCS3 mRNA in liver tissve has a significant positive relation to the level of serum leptin.[Conclusion] High expression of SOCS3 mRNA in liver may be an important mechanism for obesity leptin-resistance of DIO rat.
