摘要
目的研究细胞周期依赖性蛋白激酶1(CDK1)、三叶因子2(TFF2)在宫颈癌中的表达与人乳头瘤病毒16/18(HPV16/18)感染的相关性及其临床意义,探讨宫颈癌的发生机制。方法采用免疫组织化学检测50例宫颈癌石蜡标本、宫颈上皮内瘤变I级(CIN1)20例、CINⅡ-Ⅷ20例、正常宫颈组织20例进行CDK1、TFF2的检测,同时采用PCR技术检测HPV16/18感染情况。结果宫颈癌组织中CDK1的阳性率高于CIN和正常宫颈组织(P〈0.05),CDK1阳性表达率与病理分级有关(P〈0.05)。宫颈癌组织中TFF2的阳性率低于CIN和正常宫颈组织(P〈0.05),TFF2的阳性表达率与年龄分组有关(P〈0.05)。HPV16/18在正常宫颈组织、CIN和宫颈癌中的阳性率逐渐升高(P〈0.05),但与年龄、临床分期、病理分级、淋巴转移等均无关(P〉0.05)。CDK1和TFF2、HPV16/18的表达无相关(P〉0.05);TFF2与HPV16/18感染呈负相关(rs=-0.500,P〈0.05)。结论宫颈上皮内瘤变及宫颈癌组织中CDK1、TFF2表达的改变和HPV16/18感染共同影响CIN的发展及宫颈癌的发生和发展。
Objective To investigate the relationship between expression of CDKI, TFF2 and HPV16/18 infection in uterine cervical carcinomas. Methods Formalln-fixed, paraffin-embedded samples including 50 cases of squamous cell carcinomas, 20 cases of cervical intraepithelial neoplasia CIN Ⅱ-Ⅲ and 20 cases of CIN 1 and 20 cases of normal cervical tissues were studied. Using Polymer DetectionSystem For hnmuno-Histological Staining(PV-9000) as immunohis/o-chemical detecting method, the expression of TFF2 and CDK1 in these samples were examined. The infections of HPV type 16, 18 DNA were determined by PCR. Results There were significant differences for CDK1 between carcinomas, CIN and normal cervices, respectively ( all P 〈 0. 05). Expression of CDK1 were correlated with tumor grades ( P 〈 0. 05). The positive expression of TFF'2 in carcinomas was significantly lower than that in normal cervical tissue and CIN. There was significant differences among them ( P 〈 0. 05 ). Expression of TFF2 were correlated with age ( P 〈 0.05). The positive rate of HPV16/18was significant difference between cervical carcinomas, CIN and normal tissues ( P 〈 O. 05) , and it was not associated with age ,tumor grades, clinical stages and lymph node metastasis (all P 〉 0. 05 ). The expression of CDKI was not associated with TFF2 and HPV16/18 ( all P 〉 0. 05). The expression of HPV16/18 was associated with TFF2 ( rs = - 0. 500, P 〈 0. 05 ). Conclusions The results suggest that in tissues of CIN and cervical carcinomas in TFF2 and CDK1 expression changes and HPV16/18 infection interact with each other, which can effect the progression of CIN and carcinogenesis of cervical carcinomas.
