叶绿素铜钠盐联合环孢菌素A对免疫介导再生障碍性贫血模型小鼠的实验研究

Therapeutic effect of Sodium Copper Chlorophyllin combined with Cyclosporin A in treating AA model rats

目的探讨叶绿素铜钠盐(SCC)联合环孢菌素A(CsA)治疗再生障碍性贫血(AA)的疗效。方法建立免疫介导AA模型小鼠,分别胃饲2种不同剂量SCC12.5mg/(kg·d)+CsA12.5mg/(kg·d),SCC25mg/(kg·d)+CsA12.5mg/(kg·d),以CsA,SCC[各25mg/(kg·d)]为阳性对照组,正常对照组及模型对照组胃饲生理盐水,每组灌胃0.5ml/d,分2次给药,连续14d,分别检测血象,另取股骨作骨髓病理切片。结果SCC+CsA小剂量组WBC、HB、PLT均明显高于AA模型组,且HB,PLT分别高于SCC组和CsA组(P<0.05)。骨髓病理切片显示SCC+CsA小剂量组骨髓增生较模型组小鼠(造血组织容量<34%)改善明显(造血组织容量80%)。结论SCC+CsA促进AA模型小鼠造血功能,提高小鼠外周血白细胞、血红蛋白、血小板三系,对AA模型小鼠具有一定的治疗作用,且疗效好于单用SCC、CsA。

Objective To explore the therapeutic effect of Sodium Copper Chloropbyilin combined with Cyelosperin A in treating AA （Aplastic Anemia, AA） model mice . Methods To establish immune-mediated AA model, BALB/c rats（equal number of male and female）were randomly divided into six groups： the normal group, the AA model group, the CsA group ,the SCC group, the combination of SCC and CSA group, the combination of SCC and CSA low - dose group. They were given distilled water, CsA and SCC [Each 25mg / （kg·d）], SCC 12.5mg / （kg·d） ＋ CsA 12.5mg / （kg·d）, the SCC 25mg / （kg·d） ＋ CsA 12.5mg / （kg·d） , 0.5ml twice daily for 14 days respectively , then hemogram were tested and pathological section of femur bone marrow were observed by biopsy. Results The number of WBC,HB,PLT of the AA model mice in the combination of SCC and CsA low-does group was significantly higher than in the AA model group, the CsA group and the SCC group（ P 〈 0.05）, showing the combination of SCC and CsA low - dose group for WBC, HB, PLT recovery was better than the single - used SCC and the CsA. Bone marrow biopsy showed bone marrow hyperplasia cf the SCC Combined With CSA low- dose group compared to the AA model rats （hematopoietic tissue capacity less than 34% ） improved significantly （heznatopoietic tissue capacity 80% ）, Conclusion Sodium Copper Chlorophyllin combined with Cyehesporin A has obviose effects on recovering the hematopoiesis of the AA model rats, promoting white blood cell, hemoglobin, p｜atelet three lines. The combination of SCC and CsA low - dose group in particular haS better effect than single use of SCC and CSA.
