摘要
以拟南芥新疆生态型无菌苗真叶和叶柄为外植体,MS+6-BA 0.1 mg/mL+NAA1 mg/mL+2,4-D 1 mg/mL培养基诱导体细胞胚胎发生,采用MS+KT 1 mg/mL+2,4-D 0.2mg/mL为分化培养基,组织切片用PAS法研究体细胞胚发育过程中淀粉代谢的动态消长规律,结果表明:体细胞胚胎发育过程中出现二次淀粉积累高峰。
Somatic embryos were induced from tender leaf and leafstalk of seedling of Xinjiang ecotype of Arabidopsis thaliana.The culture medium was MS+6-BA 0.1 mg/mL+NAA 1 mg/mL+2,4-D 1 mg/mL,and the differentiation medium was MS+KT 1 mg/mL+2,4-D 0.2 mg/mL,starch metabolism was studied by PAS during the somatic embryogenesis.Results indicated that there were two peaks starch accumulation in the somatic embryogenesis of Arabidopsis thaliana.
