摘要
[目的]为桑叶总黄酮的开发利用提供依据。[方法]选取7种型号的大孔树脂进行纯化试验,利用静态吸附和动态吸附的方法,研究大孔树脂纯化桑叶总黄酮的工艺条件。[结果]D140型大孔树脂能更有效地纯化桑叶总黄酮。当上样浓度在1.041~3.122mg/ml范围内时,桑叶总黄酮的回收率较高。上样速率为1、2BV/h时,总黄酮回收率较高。上样体积为4.5BV时,树脂基本达到动态饱和吸附。当乙醇浓度为70%、洗脱速率为1BV/h时,洗脱率最大,产物纯度最高。[结论]D140型大孔树脂纯化桑叶总黄酮的最佳工艺为:以浓度约为3mg/ml的桑叶总黄酮上样,上样速率控制在2BV/h左右,上样体积为4.5BV,再用3BV、70%乙醇,以1BV/h的速率洗脱,获得的桑叶总黄酮的纯度为41.1%。
[ Objective ] The aim was to provide the basis for development and utilization of total flavonoids in Morus alba L. [ Method ] 7models of macroporous resins were selected for purification test, the technical conditions for purification of the total flavonoids in M. alba with macroporous resins was studied by using the absorption of static and dynamic state. [ Result] The maeroporous resin of Model D140 could purify the total flavonoids in M. alba much effectively. When the sampling eoncn, was within 1. 041 - 3. 122 mg/ml, the recovery rate of the total flavonoids in M. alba was higher. As the sampling flow rate was 1, 2 BV/h, the recovery rate of the total flavonoids was high, and when the sampling volume was 4.5 BV, the resin basically reached to dynamic saturated adsorption. When the ethanol conch, was 70% , and elution rate was 1 BV/h, the elution rate was the maximal and outcome purity the highest. [ Conclusion] The best technique for purification of the total flavonoids in M. alba with macroporous resin of D140 was; with the total flavonoids in M. alba at about 3 mg/ml cohen, as sampling, the sampling flow rate was controlled at 2 BV/h, sampling volume at 4.5 BV, and then, the total flavonoids was eluted by 3 BV and 70% of ethanol in rate of 1 BV/h, the 41. 1% purity of total flavonoids in M. alba could be acquired.
