摘要
目的:建立皮肤机械屏障功能障碍瘙痒模型。方法:用丙酮、乙醚混合液外擦,破坏小鼠皮肤角质层;以瘙痒行为、皮肤神经酰胺含量及角质层厚度及组织病理学改变为指标,观察造模3、5、7d后的的变化情况,以确定模型是否成功及造模的最佳时间和瘙痒行为的观察时间。结果:造模小鼠在3d、5d、7d4个时间段的瘙痒次数均高于空白组,以3d组增加最明显,而皮肤神经酰胺含量、角质层厚度及组织病理学观察以造模5d组改变最为明显。结论:用乙醚、丙酮混合液外擦可以造模皮肤机械屏障功能障碍瘙痒的模型,造模最佳时间为5 d,小鼠瘙痒行为的观察时间为30min。
Objective: To establish a skin mechanical barrier disturbance pruritus model.Methods: The mixture of acetone and ether was used to damage the keratoderma of mice by external rubbing.By taking itching,content of skin nerve acid amide,thickness of cuticle and change of histopathology as an index,the changes of the models were observed in 3,5 and 7 days to confirm the success and the best time of making the models and the time of observing itching.Results: The times of the itching of the test mice were more than that of the control group in 3,5 and 7 days.The times of the inching in the 3-day group was most obvious while the models of the content of skin nerve acid amide,thickness of cuticle and change of histopathology in the 5-day group changed most obviously.Conclusion: A skin mechanical barrier disturbance pruritus model can be made by the external rubbing with the mixture of acetone and ether.The best time of making the model is 5 days.The time to observe the itching of the mice is 30 minutes.
