摘要
为研究抑制基因nm23表达产物二磷酸核苷激酶(NDPK)在人肺癌中的表达及其意义,应用LSAB免疫组化染色方法对146例人非小细胞癌进行了分析。结果显示:nm23在人肺癌中有较高的表达,阳性率为73.23%。在鳞癌的表达随着肿瘤的增大临床分期的进展而减弱,Ⅰ+Ⅱ高于Ⅲ+Ⅳ,中~高分化的高于低分化的。在腺癌的表达中-高分化的高于低分化的。鳞癌、腺癌及腺鳞癌3组中nm23原发灶的表达水平明显高于转移灶(淋巴结),癌旁正常细支气管粘膜和肺泡上皮组织明显高于癌组织。结果表明nm23基因在肺癌的发生、发展和转移中起一定的调节作用,并可能在肺癌中作为一种抑制基因而起作用。
To determine expression of the nm23 gene products/nucleosidediphosphate kinase (NDPK) in human non-smallcell lung cancer (NSCLC) by using labelled streptavidinbiotin (LSAB) method,expression of NDPK in human lung cancer was examined in 121 cases.The results showed that expression of nm23 gene was higher in human lung cancer and 74.27% of all the specimens studied were positive staining.The expression in squamous cell carcinoma was weaken with the growth of carcinoma and the progression of PTNM stage,the expression in adenocarcinoma with moderate to full differentiation was also higher than that with poor differentiation.In the three groups of squamous cell carcinoma,adenocarcinoma and adenosquamous cell carcinoma,the expression of nm23 gene in primary lesion was higher than in metastasis lesion;level of nm23 expressed in the normal bronchiole mucosa and the alveolar epithelium adjacent to carcinoma was higher than in the carcinomatous tissue.The study indicates that nm23 gene may play a modulating role in tumerigenesis, development and metastasis of human NSCLC.It may also act as a metastatic suppressor gene in human NSCLC.
