Cloning and Sequence Analysis of Ful-length cDNA of Actin Gene from Japanese Scallop(Patinopecten yessoensis)

摘要 采用RT-PCR和RACE法从虾夷扇贝闭壳肌中分离和克隆了肌动蛋白基因cDNA全长序列。肌动蛋白基因cDNA全长1775 bp(不包括poly A),5′端非编码区94 bp,3′端非翻译区551 bp,阅读框1131 bp,编码376个氨基酸。在基因组DNA中,该基因被一个内含子分为两段,内含子位于第42和第43个氨基酸之间,长度为2041 bp。系统发育分析显示该肌动蛋白属于α类型。 The full length cDNA of actin gene of Japanese scallop ( Patinopecten yessoensis) adductor muscle was cloned and sequenced using RT-PCR and RACE methods. The cDNA was 1775 bp with 94 bp of 5UTR, 551 bp of 3UTR, and 1131 bp of ORF, which encodes 376 amino acids. One intron (2041 bp) was detected between codons 41 and codons 42 in the gene. Sequence analysis revealed that the nucleotide sequence identity of this gene between Japanese scallop and other animals was from 85% to 96%, and the similarity of amino acid sequence was from 93% to 98%. The phylogenetic analysis constructed based on the amino acid sequences suggesting that this actin be a kind of α actin.
