微核检测方法的比较研究

The Comparision of several problems in a method of micronucleus examine

本文对小鼠骨髓微核检测方法进行了对比研究。结果证明,首先在选用材料方面,股骨比胸骨好;其次在收集骨髓细胞时,冲洗骨髓腔比用止血钳挤出骨髓液取材要好;再者在选用染色方法方面,用普通的Giemsa染色比用荧光染色观察起来方便,且效果也很好。经统计学分析,实验组和阴性对照组的微核发生率分别为(7.5±1.34)‰和(2.9±0.38)‰,两组间有极显著差异(P<0.01)。

This paper compares the method of mieronucleus examine on mouse marrow cells. 20 mouse were divided randomly into experiment and negative control group. And every group has 10 mouse. Inject cyelophosphamide（60mg kg^-1 ）in intraperitoneal in experiment group. Inject sodium chloride 0.5ml in intraperitoneal in negative control group at the same time. The two groups were injected once a day and continuous 3 days. Bone marrow cells were taken out from the mice. The position of materials, the method of draw the materials and the staining aspect were compared during the experiment. And find several problems during the experiment. First, the experimental material was better choosed thighbone than breastbone. Second, it is better with washing marrow antrum to collection marrow cells than squeezeing out marrow liquid by hemostasia nip. The third,in the stain aspect, the ordinary Giemsa stain is more convenient than fluorescence stain. And the effect is very good, too. Through statistic analysis, the micronucleus incidence rate of experiment and negative group were （7.5 ±1. 34） ‰ and（ 2.9 ± 0.38 ） ‰ seperately. There is significent difference （ P 〈 0.01 ） between the two groups.
