摘要
目的选择最适合食品中沙门菌生长的增菌培养基和分离培养基,促进沙门菌生长,提高沙门菌的检出率。方法在市场上购买一批生的禽、畜类肉制品,分别通过均质器混匀,各分成两份,分别用常规培养基和本实验培养基同时增菌培养,应用改良分子信标荧光PCR检测试剂盒和mini-VIDAS全自动荧光免疫分析仪对两种增菌液进行初筛,对结果阳性者划线分离,阳性菌株经VITEK2COMPACT生化鉴定仪鉴定。结果样品经本实验增菌液增菌后,分离平板上的可疑菌落明显增多,检出沙门菌的几率也有很大提高。结论改用本实验培养基后,能有效提高沙门菌的检出率。
Objective To select a most suitable enrichment and isolating medium of Salmonella in food samples, and to promote growth of Salmonella and improve the detection rate. Methods Market meat products of poultry and livestock were collected and homogenized by a homogenizer respectively. Each sample was divided into two portions, enrichment cul- tured by the ordinary enrichment medium and the improved enrichment medium simultaneously. Modified molecular beacon based real - time POR technique and the Bio - Merieux VlDAS were used as a primary screening. The positive samples were further plate streaking cultivated. Positive clones were identified by VITEK 2compact microbiology analysis system. Results The number of suspected colonies of Salmonella in the plate that were cultivated by the improved enrichment medium was significantly higher than that from the ordinary enrichment medium. The detection rate was increased. Conclusion The improved enrichment medium can improve the detection rate of Salmonella.
