实验性SD大鼠ROU模型的建立 被引量:14

Establishing recurrent aphthous ulcer (ROU )0models of rat.

摘要 目的:利用大鼠复发性口疮(ROU)动物模型,探讨T淋巴细胞、氧自由基、微循环与ROU发病机制的关系。方法:用大鼠口腔黏膜匀浆组织加弗氏免疫佐剂注射法建立大鼠ROU模型,应用流式细胞仪检测实验前后CD4+细胞、CD8细胞的变化;分别用黄嘌呤氧化酶法和化学法检测实验前后超氧化物歧化酶(superoxide dismutase,SOD)、谷+光甘肽过氧化物酶(glutathine peroxidease,GSH-Px)和过氧化脂质分解产物(malondialdehyde,MDA)的改变;分别用生化分析法和微量毛细管法检测实验前后纤维蛋白原和红细胞压积的变化,NEX-2型板式粘度计测定实验前后血浆粘度。结果:①实验前后CD4/CD8比值下降,有显著性差异(p<0.05);②实验前后SOD下降,++GSH-Px下降,MDA升高(p<0.01);③实验前后纤维蛋白原、红细胞压积、血浆粘度均升高(p<0.01)。结论:①ROU的发生与T淋巴细胞失调有关;②证实了动物口腔溃疡黏膜的损伤系氧自由基产生所致;③成模后的大鼠存在微循环障碍。 Objective:Using the rat's recurrent oral ulcertion(ROU)models and to study directly the pathogenesis. Method:Establishing ROU models by injecting the mixture of homogenate tissue of rat's oral mucosal and immunologic adjuvant into 8 SD rats. Determing the change of CD4^+,CD8^+ by flow-cytometre;Determing the change suoeroxide dismutase(SOD),glutathine peroxidease(GSH-PX) and malondialdehyde(MDA) by the method of xanthine oxidase and chemical method respectively;Determing the changes of fibrinogen and hemat with the method of biochemistry analysis and the method of microamount capillary tube,determing the changes of plasma viscosity by NEX-2 plate caplastometer. Result:①The ratio of CD4^+/CD8^+ are obviously decreased.;②The decreasing of SOD and GSH-Px and the increasing of MDA;③In fibrinogen,hemat and plasma viscosity increase. Conclusion:①There are connection between the ROU and unbalance of T lymph cell;②Confirm ROU models by damage of free radical;③There are microcirculation obstacle in ROU models.
