摘要
目的探讨非小细胞肺癌(NSCLC)中环氧化酶-2(COX-2)和Bax的表达及其临床意义。方法采用免疫组织化学方法检测57例NSCLC癌组织(腺癌/鳞癌)、相应配对的15例正常组织及57例淋巴结COX-2蛋白和Bax蛋白的表达水平,计算其阳性百分比进行相关性分析。结果NSCLC组织中COX-2蛋白表达高于癌旁正常组织(P<0.05),腺癌高于鳞癌(P<0.05),与肿瘤组织的病理分级相关,且与淋巴结转移呈正相关(P<0.05),但与患者的年龄、性别无明显相关(P>0.05);Bax蛋白表达低于癌旁正常组织(P<0.05),腺癌高于鳞癌(P<0.05),且与淋巴结转移呈负相关(P<0.05),与年龄、性别、病理分级间差异无统计学意义(P>0.05)。结论COX-2在NSCLC尤其是肺腺癌中表达上调,Bax蛋白在NSCLC组织中表达过低,COX-2可能通过降低Bax表达,抑制细胞凋亡参与NSCLC的发生、发展。
Objective To investigate the expression of cyclooxygenase-2 (COX-2) and Bax protein in non-small cell lung cancer(NSCLC) , and its clinical significance and correlations of COX-2 and Bax. Methods Samples were obtained from 57 cases NSCLC tissues ( including adenocarcinoma and squamous cell carcinoma) , 15 sampls nomal lung tissues and 57 samples Lymphoid tissue. The expression level of COX-2 protein and Bax protein in these samples were examined by immunohistochemistry. The relationship between COX-2 and Bax were analyzed by positive percentage. Results The expression of COX-2 protein in NSCLC tissues is higher than nomal tissue which were obained nearly ( P 〈 0. 05 ), and the expression of COX-2 protein in the adenocarcinoma was higher than in the squamous cell carcinoma. That was positive related with divide degree of tumor tissues, and positive related with lymph node transfer ( P 〈 0.05 ), but was non-related with age and gender ( P 〉 0.05 ) ; The expression of Bax protein in NSCLC tissues was lower than nomal tissue which were obained neraly (P 〈 0.05) , and the expression of Bax protein in the ad was higher than in the squamous cell carcinoma( P 〈 0.05 ), it was related with negative related with lymph node transfer ( P 〈 0.05 ), but was nonrelated with age, gender, pathologic ratings ( P 〉 0.05 ). there was relationship between COX-2 and Bax ( P 〈 0. 05). Conclusions The expression of COX-2 protein is up-regulated in NSCLC tissues, particularly in adenocarcinoma. The expression of Bax proiein is over-tow in NSCLC tissues. COX-2 may participate in development and progression of NSCLC by lower expression of Bax inducing cell apoptosis.
