Na^+/H^+逆向转运蛋白NhaH短的C末端亲水域介导盐抗性和pH感应 被引量:1

摘要 Na+/H+逆向转运蛋白在维持细胞质低的Na+浓度和pH稳态等生命活动过程中扮演重要角色,然而,到目前为止,对原核微生物Na+/H+逆向转运蛋白的C末端亲水域的结构和功能还知之甚少.我们曾从达坂喜盐芽孢杆菌(Halobacillus dabanensis)D-8菌株中克隆得到第一个中度嗜盐菌来源的Na+/H+逆向转运蛋白基因nhaH.疏水性分析表明,NhaH的C末端亲水域只包含9个氨基酸残基(395PLIKKLGMI403),大大短于与之高度同源的SynNhaP1和ApNhaP的C末端亲水域.本研究采用PCR方法,构建NhaH蛋白的C末端亲水域缺失突变体nhaH-C.耐盐性实验表明,C末端亲水域的缺失显著地抑制大肠杆菌缺陷株KNabc的互补能力.基于荧光强度的翻转膜活性测定结果显示,NhaH?C的Na+/H+和Li+/H+逆向转运蛋白活性显著低于NhaH,而且前者的Na+/H+逆向转运蛋白活性向酸端偏移,表明Na+/H+逆向转运蛋白NhaH短的C末端亲水域在阳离子结合、转运和pH感应上起重要作用.
