胰岛素抵抗大鼠血管内皮功能的研究 被引量:3

摘要 目的探讨胰岛素抵抗(IR)大鼠血管内皮功能状态。方法雄性Wistar大鼠分为对照组、高脂组,喂养8w,用高胰岛素-正血糖钳夹技术稳态时葡萄糖输注率(GIR)评价IR;测定内皮型一氧化氮合酶(eNOS)及血管壁一氧化氮(N0)含量。结果GIR在两组间出现显著差异(P<0.01);高脂组大鼠血清游离脂肪酸(FFA)、甘油三酯(TG)、肿瘤坏死因子-α(TNF-α)水平高于对照组;高脂组大鼠血管内皮细胞中eNOS及NO含量明显降低(P<0.01);相关分析显示,血管壁NO含量与FFA、TNF-α呈负相关,而GIR与NO存在显著的正相关。结论IR大鼠血糖正常阶段已经存在内皮功能障碍,血中FFA、TNF-α的升高可能直接参与内皮功能障碍的发生发展。
