络脉舒通预处理对再灌注损伤心肌的影响 被引量:1

摘要 目的观察络脉舒通对兔急性心肌梗死(AMI)再灌注损伤心肌的保护作用。方法24只新西兰大白兔随机分为络脉舒通组、假手术组和对照组,每组8只。脉舒通组采用结扎冠状动脉的方法制备AMI再灌注模型;假手术组完成全部手术操作,只穿线不结扎冠状动脉。脉舒通组制模前每天灌药1次(1.5g.kg-1.d-1),连续3d,术前2h再灌药1次;假手术组和对照组均灌胃同等量生理盐水。观察各组血流动力学改变,检测丙二醛(MDA)、超氧化物歧化酶(SOD)和一氧化氮合酶(NOS)的变化,测量心肌梗死面积。结果(1)络脉舒通组缺血再灌注后2h左室收缩压(LVSP)和左心室内压最大收缩和舒张变化速率(±dp/dtmax)较对照组均显著升高(P<0.05);LVEDP则显著下降(P<0.05)。(2)络脉舒通组心肌梗死面积与对照组相比显著缩小(P<0.05)。(3)对照组MDA含量比假手术组显著增高(P<0.05),而SOD和NOS活性均显著降低(P<0.05);络脉舒通组MDA含量显著降低(P<0.05),SOD和NOS活性均显著增高(P<0.05)。结论络脉舒通通过清除氧自由基,减轻心肌再灌注损伤,获得较好的心脏保护作用。
