RP-HPLC法测定白花银背藤药材中东莨菪内酯的含量

RP-HPLC determination of scopoletin in Herba Argyreiae Seguinii

目的:建立白花银背藤中东莨菪内酯的RP-HPLC含量测定方法。方法:采用高效液相色谱法,色谱柱为LiChrospher100 RP-C_(18)(250 mm×4 mm,5μm),流动相:甲醇-0.5%醋酸水溶液(22:78),流速为1.0 mL·min^(-1),分析时间30 min,检测波长228 nm,进样体积:5～20μL。结果:该方法的线性范围为0.05～0.50 μg,r=0.9996(n=5);平均回收率(n=5)为96.9%,RSD为1.6%。结论:该法简便、准确,重复性好,可用于白花银背藤药材的质量控制。

Objective：To establish an RP - HPLC method for the determination of scopoletin in Herba Argyreiae Seguinii. Method： An HPLC method was developed. LiChrospher 100 RP - C18 column （250 mm × 4 mm, 5 μm） was adopted, and the mobile phase consisted of methanol and 0.5% acetic acid solution （22：78）. Flow rate was 1.0 mL · min^ - 1, and analysis time was 30 min. Detection was at 228 nm with injection volume of 5 - 20 μL. Results： The linear range was 0.05 - 0.50 μg, r = 0. 9996 （ n = 5 ）, and the average recovery （ n = 5 ） was 96.9%, RSD was 1.6%. Conclusion：The method is simple,accurate and reproducible,suitable for the quality control of scopoletin.
