摘要 目的:建立金银花药材的HPLC特征图谱,为金银花药材的质量鉴定提供依据。方法:采用Phnomenex Gemini C_(18)色谱柱(250 mm×4.6 mm,5μm),以0.5%磷酸-乙腈为流动相梯度洗脱,流速为1 mL·min^(-1),柱温40℃。检测器为紫外检测器和电化学检测器。结果:得到了分离度较好的HPLC-UV(238 nm)和HPLC-ECD(250 mV)2种特征图谱,两者具有良好的相关性和互补性。分别识别了17个和9个特征共有峰。结论:该方法稳定、准确、可靠,可用于金银花药材的质量控制。 Objective:To establish the HPLC specific chromatograms of Flos Lonicerae Japonicae, it may provide the basis for identification for quality of Flos Lonicerae Japonicae. Methods: Chromatographic analysis was achieved on a Phnomenex Gemini C18 column(250 mm ×4. 6 mm,5 μm) using a mobile phase of 0. 5% phosphoric acidacetonitrile eluted with a linear gradient. The detectors were consisted of UV and ECD detector. The flow rate was maintained at 1.0 mL· min^-1, with the column temperature at 40℃. Results:The method provided two kinds of specific chromatogram with excellent separation, which were HPLC - UV ( 238 nm) and HPLC - ECD ( 250 mV) specific chromatograms. They had a good correlation and complementation with each other. Seventeen common peaks and nine common peaks were confirmed in each specific chromatogram, respectively. Conclusion:The method is stable, accurate and creditable. It can be used for the quality control of Flos Lonicerae Japonicae.
