摘要 以菜豆为原料,对菜豆皂苷的提取和纯化工艺条件进行研究。通过对比实验,确定了提取剂为乙醇溶液;根据单因素实验结果进行正交实验,结果表明,最佳提取工艺条件为:提取温度70℃、提取时间30min、乙醇浓度90%、料液比1:1.5;最佳纯化工艺条件为:pH8.0、乙醇浓度90%、解吸流速30滴/min、解吸时间1.0h。在最优提取及纯化条件下,菜豆皂苷的得率和纯化率分别为0.286%、58.75%。 The extraction conditions and purification method of Phaseolus vulgaris saponin was studied using Phaseolus vulgaris as raw materials. The extraction agent was ethanol solution, which was determined after ample comparing. The orthogonal experiment was adopted according to the results of the single-factor test. The result indicated that the optimum conditions of extraction were as follows: extract temperature was 70℃, the time of extraction was 30min, ethanol concentration was 90%, liquid ratio was 1 : 1.5. The optimum conditions of the purification was as follows= pH8.0, 90% ethanol concentration, desorption velocity was 30drip/min, desorption time 1.0h. The extraction yield under the optimum conditions for the Phaseolus vulgaris saponin was 0.286% and the purification yield was 58.75%.
