摘要
运用反相高效液相色谱法测定了人血清中黄连素浓度。以改性甲醇为流动相,检测波长为347nm,外标法定量,线性范围为0.2~2.0mg/L(r=0.9996),平均回收率为90.73%,最低检测限2.55ng,日内、日间误差均小于8%。方法灵敏、准确、快速。
Berberine is a safe and effcetive anti arrhythmic drug. We do not clearly know about the dose effect relationship. Besides, berberine is not fit for administration by i.m. and i.v.. Therefore, it is necessary to build a PD model of berberine, study its pharmacodynamics parameters in vivo, and provide a concrete criteria in the clinical usage. In this article, we set up a method for determination of concentration of berberine in human serum by RP HPLC with Lichrosorb RP 18 column (250mm×4mm) and mobile phase of MeOH containing 015% friethylamine. The content of berberine was determined by the external standard method with UV detector at 347nm. The linear range was 02 2mg/L (r=09996). The detectable limit was 255ng. The average recovery was 9073% and the coefficients of variation were below 80%. The method is sensitive and accurate.
