摘要
目的通过主动脉根部注射兔的自体骨骼肌细胞悬液和VEGF、HGF因子,观察对急性缺血/再灌注损伤的心脏功能的影响。方法取42只成年兔,随机分成六组,培养液对照组(A)、细胞移植组(B)、VEGF因子组(C)、VEGF因子+细胞移植组(D)、HGF因子组(E)、HGF因子+细胞移植组(F)。取臀肌进行骨骼肌细胞的体外分离,制成相应的细胞移植液。结扎兔的左冠状动脉制成急性心肌梗死再灌注模型,通过主动脉的根部分别给予移植液注射。移植术4周后,通过心脏超声对各组实验兔心功能进行检查、记录。对六组兔梗死心肌及周围组织、梗死的心室壁厚度进行HE染色情况、myosin免疫组化、心动超声指标进行观察和统计学分析。结果在B、D、F移植组心功能明显好于A、C、E组,B、C、D、E、F组好于A组,心动超声指标进行比较,差异有统计学意义(P<0.05)。结论通过主动脉根部注射骨骼肌细胞和VEGF、HGF因子可以提高兔心肌缺血/再灌注损伤后心功能状态,降低损伤心肌的纤维化。
Objective To study the effect of the function of myocardial ischemia reperfusion injury by the base of the aorta rout to transplanted the autologous skeletal muscle cell and VEGF or HGF factor into the injury myocardium. Metheds 42 rabbits were randomly divided into six groups, cell group ( B ) and the control( A ) , cell + VEGF group ( D ) and the control ( C ) , cell + HGF group ( F ) and the control ( E). The autologous muscle cell isolated from their buttocks muscles , and were made skeletal muscle cell suspension, skeletal muscle cell + VEGF suspension, skeletal muscle cell + HGF suspension . The left coronary arteries of the adult rabbits were ligated to make myocardial ischemia reperfused models, then all suspensions were injected into the base of aorta slowly of the experimental groups individually. Culture medium, cuhure medium of VEGF factor, culture medium of HGF factor were reperfused into the three control experimental group individually. The hearts were observed with Eehocardiography after four weeks. The myocardium were harvested and observed under a light microscope after stained by HE. Then myosin immunohistochemistry were conducted. Result The function of hearts of B,D,F group were better than other groups significantly. The function of hearts of VEGF and HGF group were better than the A group. Conclusion The transplanting skeletal muscle cells abstracted directly can enhanced cardiac function after myocardial ischemia reperfuse injury by using intra - aorta route. VEGF factor and HGF factor injection can enhanced cardiac function 'after myocardial injury, meantime reduce infarcted myocardium fibriaustroply.
