摘要
目的探讨利用RNA干扰技术在裸鼠体内抑制survivin基因表达并诱导前列腺癌细胞凋亡的作用。方法设计、构建靶向survivin的重组质粒载体并导入裸鼠皮下移植瘤,称量瘤重,计算抑瘤率,应用RT-PCR法检测survivin基因的干涉效率,TUNEL染色法技术检测细胞凋亡效果。结果PBSsi-survivin A和PBSsisurvivin B两种阳性表达载体转染组均具有显著低瘤重和高抑瘤率特点,两组均显著抑制了Survivin mRNA表达,并诱导前列腺癌细胞凋亡。结论RNAi可有效抑制前列腺癌PC-3细胞survivin基因mRNA表达,减缓肿瘤生长,并诱导细胞凋亡。
Objective To investigate the effect of inhibition of survivin expression in prostate cancer cell line PC-3 by RNAi on the apoptosis of prostate cancer PC-3 cells. Methods siRNA expression vectors of surviving were constructed and transfected into prostate cancer cell line PC-3 cells, then planted in subcutaneous of nude mouse. The weight of tumor was weighed. Inhibition of surviving was assessed by RT-PCR and cell apoptosis was detected by TUNEL staining. Results Expression vectors PBSsi-survivin A and PBSsi-survivin B were found to inhibit the expression of surviving and enhance cell apoptosis, the weight of tumor were also found to decrease. Conclusion The surviving RNAi can effetively inhibit the mRNA expression of survivin in prostate cancer cell line PC-3 and reduce the growth of tumor and induce tumor cell apoptosis.
