摘要
目的探讨刺五加注射液体外诱导大鼠骨髓基质细胞分化为神经样细胞的最佳浓度。方法体外密度梯度法分离培养骨髓基质细胞,以不同浓度刺五加注射液对其进行体外诱导,用MTT法检测各组细胞生长情况,用免疫细胞化学方法计算各组神经样细胞阳性数加以比较。结果用不同浓度刺五加注射液诱导后均可表现为典型的神经样细胞形态,其中0.3mg/ml组和0.4mg/ml组细胞生长情况与免疫组化结果均与其余各组差异有统计学意义(P<0.05或P<0.01)。结论刺五加注射液体外诱导成年大鼠骨髓基质细胞分化为神经样细胞以0.3~0.4mg/ml浓度最佳。
Objective To study the optimal concentration o5 Ciwujia Injection for rat bone marrow stromal cells differentiating into neuron-like cells in vitro. Methods The density-gradient technique was used to separate and culture the bone marrow stromal ceils and the different concentration of Ciwujia Injection was applied for inducement in vitro. The growing of cells of all groups was determined with MTT and the neuron-like cells of each group were calculated by immunocytochemistry examinauon for comparison. Results With inducement of Ciwujia Injection in different concentrations, the cells of all groups were typically shaped like neurons. Of them, the growing of cells in 0. 3mg/ ml and 0. 4mg/ml groups and their immunocytoehemistry results were both showing differences from other groups in statistics (P〈0.05 or P〈0.01). Conclusion The concentration of 0. 3〈0.4mg/ml was optimal for inducement of bone marrow stromal ceils differentiating into neuron-like ceils in vitro in adult rats.
