瑞芬太尼对慢性坐骨神经损伤大鼠T细胞增殖及NK细胞活性的影响 被引量:1

EFFECT OF REMIFENTANIL ON T CELL PROLIFERATION AND NK CELL ACTIVITY IN RATS WITH CHRONIC CONSTRICTION INJURY OF SCIATIC NERVE

摘要 目的:研究瑞芬太尼对慢性坐骨神经持续压迫损伤(chronic constriction injury,CCI)大鼠的T细胞增殖及自然杀伤细胞(NK细胞)活性的影响。方法:40只雄性W istar大鼠分为实验组、盐水对照组和假手术组,用手术方法结扎坐骨神经制作疼痛模型,观察大鼠缩腿反应潜伏期(paw with-drawal latency,PWL)。同时分别应用乳酸脱氢酶释放法和刀豆蛋白A(Con-A)刺激淋巴细胞转化法测定NK细胞活性和T细胞的增殖。结果:实验组大鼠出现PWL增高、T淋巴细胞功能和NK细胞活性抑制,与对照组相比有显著性差异(P<0.05)。结论:瑞芬太尼治疗能够对CCI大鼠痛敏反应产生剂量依赖性抑制作用,但大剂量应用会抑制NK细胞活性和T细胞增殖反应。 Objective: To investigate the effects of remifentanil treatment on T-lymphocyte proliferation and natural killer (NK) cell activity in rats receiving chronic constriction injury (CCI) of sciatic nerve. Methods : 40 male Wistar rats were separated into three groups of experimental group, saline control group and sham operation group. Except sham operation group, all the other rats were operated and given ischiadic nerve ligation. The paw withdrawal latency (PWL) and the proliferation of T lymphocytes were ob- served by lactate dehydrogenase releasing method, the activity of NK cells was measured by Con-A stimulating lymphocytes transformation method respectively. Results: NK cell activity and T cells proliferation in experimental group were significantly suppressed compared to control group (P 〈 0.05 ). Conclusion : These data suggest that remifentanil treatment can suppress thermal hyperalgesia in a dose dependent manner in rats with sciatic CCI, while it can inhibit NK cell activity and T cell proliferation with high dose.
