开窍中药对牛肾上腺髓质细胞儿茶酚胺分泌的影响 被引量:16

Effects of Traditional Chinese Medicines for Inducing Resuscitation on Catecholamine Secretion in Bovine Adrenal Medullary Cells

摘要 目的探讨开窍中药冰片、牛黄、麝香、石菖蒲及苏合香对牛肾上腺髓质细胞儿茶酚胺分泌的影响。方法体外培养牛肾上腺髓质细胞,HPLC-ECD检测其分泌儿茶酚胺的含量,分别考察5种开窍药对乙酰胆碱、藜芦定碱和56 mmol·L-1高钾溶液诱导的儿茶酚胺分泌的影响。结果5种开窍中药均显著抑制乙酰胆碱、藜芦定碱诱导的儿茶酚胺分泌(P<0.05或P<0.01)而对高钾溶液诱导的儿茶酚胺分泌均无显著影响;对乙酰胆碱诱导的儿茶酚胺分泌的抑制作用随浓度升高而增强(P<0.05或P<0.01)。结论开窍中药具有抑制交感活性作用,且通过乙酰胆碱受体途径和电压依赖型钠离子通道发挥作用。 OBJECTIVE To investigate the effects of traditional Chinese medicine (TCM) for inducing resuscitation include boineol, cow-bezoar, musk, grassleaf sweetflag rhizome and storax on catecholamine (CA) secretion from cultured bovine adrenal medullary cells. METHODS Bovine adrenal medullary cells were primary cultured at a density of 5×10^5 cells/well (24 wells plates, costar), which were used in 3 -7 d. The cells were incubated with acetylcholine, veratridine and 56 mmol·L^-1 K^+ solution in the presence or absence of the above five drugs for 10 min. Then the contents of catecholamine secreted into the supernatants from the cultured cells were determined by HPLC-ECD analysis. RESULTS Incubation of cells with all the indicated drugs for 10 min caused a significant decrease in CA secretion induced by acetylcholine(300 μmol·L^-1 ) and veratridine ( 100 μmol·L^-1 ) ( P 〈 0.05 or P〈0. 01 ), while no effect was observed for 56 mmol·L^-1 K^+ induced CA secretion. The inhibition of ACh-induced CA secretion from bovine adrenal medullary cells showed a dose-dependent manner. CONCLUSION These results suggest that drugs for inducing resuscitation inhibit the activities of sympathesis via nicotinic acetylcholine receptor-ion channel pathway and voltagedependent Na^+ channel pathway in adrenal medullary cells.
