摘要
目的:测定急性出血坏死性胰腺炎(AHNP)大鼠血浆及胰腺组织内皮素(ET)含量变化,探讨其在发病机理中的作用。方法:5%牛磺胆酸钠溶液胆胰管注入,诱发大鼠AHNP。动态测定血浆及胰腺组织内皮素(ET)、胰腺血流量(PBF)、肌醇三磷酸(IP3)和胰腺细胞[Ca2+]i的变化,胰腺组织病理评分(PM)定量分析。结果:AHNP大鼠血浆及胰组织ET含量早期显著增加,血浆ET初始升高滞后于胰组织ET,两者间及两者与PBF、IP3、[Ca2+]i、PM呈显著相关性,但胰组织ET相关更密切。结论:胰组织ET可能介导了胰腺早期缺血,血浆ET进一步加重AHNP大鼠的病理生理改变。
Objective: To measure the content of endothelin (ET)in the plasma and pancreas in rats with acute hemorrhagic necrotizing pancreatitis (AHNP) and determine the role of ET in the pathogenesis of AHNP. Methods: After the model of AHNP was established with the injection of 5% sodium taurocholate into the bilepancreatic duct in rats, the changes of pancreatic blood flow (PBF), levels of ET and 1, 4, 5trisphosphate (IP3) in pancreatic tissue and the content of [Ca2+]i in pancreatic cells were dynamically observed. Meanwhile, the pathological changes of the pancreatic tissue (PCPT) were also quantitatively analyzed. Results: The level of ET in the plasma and pancreatic tissue was significantly increased and its increase in the plasma was slower than that in the pancreatic tissue in the early stage. Significant correlation was found among ET level in the plasma, ET level in the pancreatic tissue, PBF, IP3, [Ca2+]i and PCPT and the first was more closely correlated with the last four than the second. Conclusion: ET in the pancreas might mediate pancreatic ischemia in the early stage and that in the plasma aggravate pancreatic injury after AHNP in rats.
