摘要
应用凝胶阻滞技术(gelretaldation),在甲基硝基亚硝胍(MNNG)诱发的遗传不稳定的Vero细胞(非洲绿猴肾细胞)抽提物中,发现存在特异性针对G·T的错配结合蛋白(mismatchbindingprotein),能选择性识别DNA中的G·T错配,并与之结合.经与正常Vero细胞相比较,证实其功能并无缺陷。从而排除了MNNG通过对错配结合蛋白功能的损伤而诱发细胞遗传不稳定的可能机制。
Mismatch binding protein specific for G.T was identified by using gel retardation technique in the extracts of genetically instable monkey kidney Vero cells induced by MNNG (N-methyl- N' - nitro - N -nitrosoguanidine) .The protein could bind selectively to G.T mismatched in DNA and no functional defeciency of the protein was fed in the above extracts ,ascompared with the normal one . So that the possible mechanism that defeciency of mismatch binding protein participated in the inducement of Vero cell genetic instability by MnNG could be exclude .
