摘要
应用骨髓细胞长期培养方法动态观察10例AML贴壁细胞生长特点,探讨了AML基质细胞在白血病发生,发展中的作用,发现5/10例贴壁层较早形成完全融合,培养~3周各例AML贴壁层出现一些大型多角扁平细胞,其中2例M5出现一些大型巨噬细胞,培养至8~9周,6/10例AML融合消失,贴壁层仅留下一种大型多角扁平细胞和大量由其介导获独立生长的造血细胞(为幼稚细胞)表明这些大型基质细胞功能较为活跃。
The growth characteristic of the adherent cells in long - term marrow culture of 10 cases with acute myeloid leukemia(AML) was observed in oafer to undersed the effects of stromal population on the development of leukemia. An early complete confluence of the adherent layer in 5/10 AMLs was noticed. During the first 2 ~ 3 weeks of culture, there were some lug flat- angulated cells in the adherent cells in all samples. Some giant macrophages were found in the adherent layer in 2/10 cultures of acute monocytic leukemi a. In the 8 ~ 9 week - old - culture, the phenomena of conlluence disappend in the adherent layer in 6/10 samples and only a kind of stromal cell, large flat - angUlated cells which can support leukemic cells to grow and to make some leukemic cells acquire the ability of self- maintain, was retained in the adherent layer.This demonstrated that the alteration of the component in marrow stromal cells might be one of reasons that the proliferation and differential direction of hematopoietic cells was influenced. Excessive growth of some stromal cells may be one of the reasons of occurence and development of acute myeloid leukemic.
