腺苷预处理对大鼠局灶性脑缺血再灌注损伤后胶质源性神经营养因子表达的影响 被引量:4

Influence of adenosine preconditioning on GDNF with local cerebral ischemia - reperfusion injury in rats

摘要 目的观察腺苷预处理对大鼠局灶性脑缺血再灌注后胶质源性神经营养因子表达的影响。方法制作大鼠大脑中动脉缺血2h后再灌注损伤模型。SD大鼠60只,随机分为假手术组(F组)、缺血再灌注组(IR组)、腺苷预处理组(AP组),依术后处死动物时间的不同,每组再分为4个亚组2、6、24、72h组(F26,24,72h,IR2、6、24、72h,AP2、6、24、72h),每个亚组5只动物。结果用免疫组化法检测脑缺血再灌注组织中胶质源性神经营养因子(GDNF)表达。结果F组GDNF阳性细胞表达极弱,IR组和AP组在脑缺血再灌注后2h在缺血半暗带周围出现GDNF弱阳性表达,6h后GDNF阳性细胞表达增多,24h GDNF阳性细胞表达达到高峰,72hGDNF阳性细胞表达开始减少。AP各亚组GDNF阳性细胞数与F和IR各相应亚组GDNF阳性细胞数比较差异有统计学意义(P〈0.05),IR各亚组GDNF阳性细胞数与F各相应亚组GDNF阳性细胞数比较差异有统计学意义(P〈0.05)。结论腺苷预处理能显著增强大鼠局灶性脑缺血再灌注损伤后神经细胞的蛋白合成和分泌功能、增加脑组织中胶质细胞源性神经营养因子的表达。 Objective To observe the influence of adenosine preconditioning on the expression of glail derived neurotrophic factor with local cerebral ischemia - reperfusion injury in rats. Methods The model of reperfusion after 2 hours MCAO ( middle cerebral artery occlusion) was made. 60 Sprague -Dewley rats were randomly divided into the false operated group(F group, n =20) ,the ischemia-reperfusion group (IR group, n =20) and the adenosine preconditioning group (AP group, n =20) F, IR and AP groups were further divided into four subgroups respectively ( F2 h ,F6 h ,F24 h ,Fn h ;IR2 h ,IR6 h ,IR24 h ,IRn h ; AP2 h ,AP6 h, AP24 h ,APn h ) by the time of killing the rats, with 5 rats in each subgroup. The expression of glail derived neurotrophie factor (GDNF)was detected by the immunohistochemical method. Results The expression of GDNF masculine cells was very weak F group. The expression of GDNF mascu- line cells was weak in 2 h subgroup, reinforcing in 6 h subgroup, peak time in 24 h subgroup and weaken in 72 h subgroup in both IR and AP group. The number of GDNF masculine cells in each subgroup of AP group was more than those in F and IR according subgroup (P〈0.05). The number of GDNF masculine cells in each subgroup of IR group was more than those in F according subgroup (P〈0.05). Conclusion Adenosine preconditioning can obviously strengthen the composing and secreting function of nerve cells,increase the expression of GDNF masculine cells in the brain of rats after brain ischemia-reperfusion.
