摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)中ISG15 mRNA表达及其与疾病活动性的关系。方法采用逆转录实时荧光定量聚合酶链反应(FQ-RT-PCR)的方法,在Light Cycler Real time PCR检测仪上检测89例SLE患者(活动期50例、缓解期39例)、结缔组织病(CTD)患者34例(疾病对照组)、健康体检者35名(健康对照组)外周血PBMCs中ISG15 mRNA的表达水平和含量。以ΔCt=Ct(待测基因)-Ct(内参基因)来比较基因表达水平的变化。结果活动期SLE患者ISG15 mRNA表达水平(ΔCt=2.04±1.07)明显高于缓解期SLE患者(ΔCt=5.48±1.72,t=11.572,P<0.01);活动期SLE患者ISG15 mRNA表达水平(ΔCt=2.04±1.07)明显高于疾病对照组(ΔCt=5.90±1.72,t=12.69,P<0.01)和正常对照组(ΔCt=5.66±1.63,t=12.359,P<0.01);缓解期SLE患者ISG15 mRNA表达水平与正常人及疾病对照组的差异无显著性(P>0.05)。SLE患者PBMCs中ISG15 mRNA的表达水平与SLE疾病活动评分指数(SLEDAI)评分及抗双链DNA抗体呈显著正相关(均P<0.01),与补体C3、C4呈显著负相关(均P<0.01)。结论活动期SLE患者ISG15 mRNA表达水平显著高于正常人、疾病对照组和疾病缓解期,提示ISG15的表达可能参与了SLE的发病过程并与疾病的活动性显著相关。
Objective To detect the expression of ISG15 mRNA in peripheral blood mononuclear cells(PBMCs)in patients with systemic lupus erythematosus(SLE), and explore its relationstfip with activity of SLE. Methods Reverse transcription-real time quantitative polymerase chain reaction(FQ-RT-PCR) was used to examine ISG15 mR.NA expression in peripheral blood monocytes of 50 eases of SLE in active stage,39 eases in stable stage,34 cases in CTD,and 35 healthy controls.Results The mean levels of ISG15 mRNA expression of SLE inactive stage( △Ct = 2.04 ± 1.07 ) was higher than that of stable stage( ACt = 5.48±1. 72, t = 11.572, P 〈 0.01), CTD(△Ct = 5.90±1.72, t = 12.69, P 〈 0.01 ), and healthy controls(△Ct = 5.66 ±1.63, t = 12. 359, P 〈 0.01 ) .There was no significant difference of expression levels of ISG15 mRNA among stable stage SLE patients, CTD diseases patients and healthy controls( P 〉 0.05) .The expression levels of ISG15 mRNA in PBMCs with SLE were correlated significantly with the SLEDAI scores ( P 〈 0.01 ), anti-dsDNA( P 〈 0.01 ), complement C3 ( P 〈 0.01 )and complement Ca ( P 〈 0.01 ). Conclusion Tiffs study indicated that the ISG15 mRNA expression levels of PBMCs is significandy elevated in patients with active SLE,it maybe correlated to pathogen- esis and activity of disease.
