S-生物丙烯菊酯处理后人淋巴细胞基因表达谱的变化

摘要 目的运用基因芯片技术研究S-生物丙烯菊酯对正常人淋巴细胞基因表达谱的变化。方法取正常人外周血淋巴细胞,加入S-生物丙烯菊酯刺激,提取总RNA与Agilent Human1B寡核苷酸基因芯片杂交,研究基因表达谱变化。结果正常淋巴细胞在S-生物丙烯菊酯刺激后共有346个基因表达发生变化,其中16个免疫和防御相关基因均表达下调,6个凋亡相关基因表达上调,1个凋亡相关基因表达下调。结论S-生物丙烯菊酯处理后淋巴细胞基因表达谱的总体变化趋势是部分免疫应答基因表达下调,促进凋亡,抑制增殖。 Objective To explore the effect of S-bioallethrin on the gene expression profile of human lymphocytes. Methods Total RNA were extracted from peripheral blood lymphocytes treated by S-bioallethrin and hybridized with the Agilent human 1B 60mer oligonucleotide microarray, and then the changes of gene expression profile were studied. Results Changes of gene expres- sion in 346 genes were found in human lymphocytes treated with S-bioallethrin,of which, 16 genes related to immunity and defense and 1 gene related to apoptosis were down-regulated,and 6 genes related to apoptosis were up-regulated. Conclusion The total tendency of gene expression of human lymphocytes treated with S-bioallethrin is that some genes related to immunity and defense are down-regulated for promoting apoptosis and inhibiting proliferation.
