摘要
目的研究雌激素在体外对正常人精子运动参数和胞内钙离子的影响。方法用不同浓度的雌激素类物质17β-雌二醇(E_2)作用于人精子,以汁算机辅助精子分析系统(CASA)观察人精子体外运动参数的变化;用腺昔酸环化酶抑制剂预处理人精子,通过流式细胞术检测E_2、非透膜性大分子物质雌二醇-牛血清白蛋白复合物(E_2-BSA)对精子胞内Ca^(2+)浓度的影响。结果0.1μmol/L的E_2可显著提高精子前向运动百分率、平均曲线运动速度、平均直线运动速度和平均路径速度(P<0.05),10μmol/L的E_2明显降低精子的前向运动百分率、平均曲线运动速度和平均路径速度(P<0.05),0.0lμmol/L的E_2对精子的各项运动参数无显著影响;E_2(0.1μmol/L)、E_2-BSA(5μmol/L)作用后,精子胞内Ca^(2+)浓度均显著升高(P<0.05);腺苷酸环化酶抑制剂可明显抑制E_2或E_2-BSA引起的精子胞内Ca^(2+)浓度的升高(P<0.05)。结论雌激素在体外可增强正常人精子的运动力,该效应可能是通过雌激素与人精子膜上雌激素受体结合后,升高精子胞内Ca^(2+)的浓度以及激活腺苷酸环化酶而实现的。
Objective To study the effects of estrogen on sperm movement parameters and intracellular calcium concentrations ( [ Ca^2+ ] i) in fertile men in vitro. Methods Sperm movement parameters were determined by computer assisted sperm analysis (CASA) in the presence of different concentrations of E2. Effects of adenylyl cyclase inhibitor and conjugated E2(E2-BSA) on intracellular calcium concentrations ( [ Ca2 + ] i) in sperms were respectively measured by flow cytometry. Results E2 at concentration of 0.1 μmol/L could significantly improve the sperm progressive motility, VSL, VCL and VAP( P 〈 0.05). E2 at concentration of 10μmoL/L evidently reduced the progressive motility, VSL, VCL and VAP ( P 〈 0.05). And E2 at concentration of 0. 01 μmol/L had no effect on sperm movement parameters. Both E2(0.1 μmol/L)and E2-BSA( 5μmol/L) significantly increased the [Ca^2+ ]i in sperms. The adenylyl cyclase inhibitor could inhibit the increase of [Ca^2+ ]i induced by E2 or E2-BSA ( P 〈 0.05). Conclusions Estrogen can promote the sperm motility in fertile men. The mechanism may be explained by the [ Ca^2+ ]i increase and activation of adenylyl cyclasc caused by the integration of estrogen with the membrane receptor on human sperm.
