通过原生质体质粒转化提高Streptomyces actuosus诺西肽产量的研究 被引量:3

摘要 研究利用大肠杆菌-链霉菌穿梭质粒pSE34,构建vhb基因的重组质粒pSEVhb,采用原生质体质粒转化法,将携带vhb的重组质粒转入诺西肽产生菌Streptomyces actuosus中,获得重组菌S.actuosus/pSEVhb。通过CO结合试验,证明VHb在重组菌中成功表达。限氧试验表明,重组菌S.ac-tuosus/pSEVhb诺西肽的的产量明显高于出发菌株。传代试验重组菌稳定,适合工业化大生产。 VitreoseiUa hemoglobin (VHb) gene was cloned into vectors containing erm promoter and was introduced into Streptomyces actuosus by protoplast-plasmid transformation. The successful expression of VHb gene was verified by the analysis of carbon monoxide-binding difference spectra. Through the oxygen-limited experiments, the output of nosiheptide of the recombinant S. actuosus/pSEVhb was higher than that of the original strain. The recombinant strain was stable and suitable for the industrial application.
