摘要
设计并合成了一种新的配体水杨醛缩肼基甲酸甲酯(salicylidenehydrazinocarboxylate,HL),并以乙醇为溶剂,回流条件下合成了HL与铜的配合物。对其结构进行元素分析、摩尔电导率、紫外吸收光谱、红外吸收光谱、荧光光谱及电化学表征,结果表明,配合物同时含有2个M─O和1个M─N键形成的五元环和六元环,HL为多齿配体。并用荧光光谱法和电化学方法对配合物与DNA的相互作用进行了研究,结果表明,配合物与DNA的相互作用为插入式结合,其结合常数为3.40×105L.mol-1,该配合物与ssDNA和ds-DNA有不同的荧光光谱性质,利用配合物的这种性质可以将其作为探针试剂识别dsDNA和ssDNA。
A novel Schiff base ligand salicylidenehydrazinocarboxylate(HL) and its copper complex were synthesized and their structures were characterized by elemental analysis, molar conductivity, UV and IR spectrum analysis, fluorescence spectrometric analysis and electrochemical method, respectively. Experimental results showed that the CuL complex had five-membered and six-membered rings containing two M-O, and one M-N bonds and that the ligand was of a multidentate one. The interaction of the CuL complex with DNA was studied with fluorescence spectrometry and electrochemical method. The results indicated that CuL complex interacted with DNA by intercalation with a binding constant of 3.40× 10^5 L·mol^-1. It was also found that the characteristics of fluorescence spectrum of dsDNA-CuL complex was quite different from that of ssDNA-CuL complex, thus the CuL complex can be used as a probe to discriminate dsDNA and ssDNA.
