摘要
采用毛细管区带电泳(CZE)技术,研究了盐酸普罗帕酮(propafenone hydrochloride)与人血清白蛋白(HSA)的结合作用机制。在以硼砂-碳酸氢钠(pH10,50 mmol/L)为运行缓冲溶液,运行电压17 kV,进样时间12 s,紫外检测器(214 nm)的条件下检测,结合常数和结合位点数在298 K和310 K分别为K298K=2.24×107L.mol-1,n298K=1.2及K310K=3.12×107L.mol-1,n310K=1.3。同时运用荧光光谱研究了盐酸普罗帕酮与人血清白蛋白的结合作用机制;并从热力学参数推得了药物分子与人血清白蛋白分子间的作用力类型等分子间的相互作用信息。
The binding interaction between propafenone hydrocloride and human serum albumin (HSA) was studied by capillary zone electrophoresis method (CZE) with UV detection. The UV wavelength and the applied voltage were set at 214 nm and 17 kV, respectively. All of the separa- tions were performed in 50 mmol/L Na2B4O7 -NaHCO3 (pH 10)buffer solution. Under optimal conditions, the binding constant and the number of binding site at 298 K were 2.24×10^7 L·mol^-1 and 1.2, respectively; and at 310 K, 3.12×10^7 L·mol^-1 and 1.3, respectively. The binding mechanism was also studied with fluorescence spectrometry. Based on the thermodynamic parameters, the type of the main binding force between propafenone hydroeloride and HSA was obtained.
