摘要
目的:体外筛选海洋真菌多糖(YCP)结合的免疫细胞类型。方法:采用化学偶联的方法将荧光胺(FLA)和YCP结合,制备荧光探针YCP-FLA。分离不同的免疫细胞(巨噬细胞、单核细胞、Kupffer细胞等)与YCP-FLA共孵育,荧光显微镜观察以及流式细胞仪检测YCP-FLA和不同免疫细胞结合特性。结果和结论:YCP-FLA可以直接与单核巨噬细胞(腹腔巨噬细胞、脾巨噬细胞、肝Kupffer细胞和人外周血单核细胞)结合,推测YCP可能是通过单核/巨噬细胞表面的受体,调节免疫活性。
Aim: To screen the immunoeytes which could bound to YCP in vitro. Methods: Fluorescence-labeled YCP(YCP-FLA) was prepared by chemical coupling to bind fluoreseamine with YCP. Immunoeytes of macrophage, monocytes, Kupffer cells and hepatic cells were separated and incubated with YCP-FLA. Fluorescent microscope and fluorescent activated cell sorting (FACS) were used to analyze the binding of YCP-FLA to immunoeytes. Results and Conclusion: YCP-FLA could bind to monoeyte maerophage (peritoneal maerophages, macrophage in spleen, monocytes in peripheral blood and Kupffer cells) directly. It is assumed presume that YCP can bind to receptors of monocyte macrophage and mediates the related immunomodulation.
