银杏叶提物对肝癌细胞生长株、甲胎蛋白分泌及γ-GT活性的影响

摘要 目的研究银杏叶提物(EGb)对肝癌细胞生长、甲胎蛋白(AFP)分泌及γ-谷氨酰转移酶(γ-GT)活性的影响。方法以两种人肝癌细胞株SMMC-7721细胞和Bel-7402细胞分别与EGb(17.5~105mg/d)共培养6d,利用MTT比色、ELISA及细胞分化检测等方法。结果Egb(17.5、35、70、105mg/d)均出现细胞增殖抑制,亚细胞结构趋于正常,并且明显降低HepG2细胞高水平分泌AFP。EGb(35、70mg/d)可抑制肝SMMC-7721细胞的γ-GT活性,升高白蛋白(ALB)含量。EGb(70、105mg/d)亦可抑制肝癌Bel-7402细胞的γ-GT活性,并升高ALB含量。结论EGb具有抑制肝癌生长、AFP分泌及γ-GT活性的作用,诱导其在体外向正常肝细胞分化。 Objective To study effecttiontion of hepatoma cell line growth stock、α-FP excretion and γ-glutamyl transferase activity by applying ginkgo biloba leaves extract(EGb).Methods To discern Vitamin D anthropo-hepatoma carcinoma cell line growth stock SMMC-7721 cell and Bel-7402 cell,to co-culture 6 days uniting EGb(17.5~105 mg/d),to utilize(Mono-nuclear cell direc cytotoxicity assay)MTT shade selection,(enzyme-linked immunosorbentassay)ELISA and cellular differentiation detecting system.Results As all to show cytostasis,subcellular structure to normalin,and obviously step down HepG2 cell homo-level secrete α-FP,using EGb(17.5、35、70、105 mg/d).Egb(35、70 mg/d)hold-back SMMC-7721 hepat-cellularγ glutamyl transferase activity,to heighten albumin contents.Eg(70、105 mg/d)hold-back Bel-7402 hepat-cellularγ glutamyl transferase activity,to heighten albumin contents.Conclusion EGb have effection which to inhibit hepatoma cell line growth stock、α-FP excretion and γ-glutamyl transferase activity;to guide to hepat-cellular differentiation in vitro.
