高糖及AGE对高压下THP-1源泡沫细胞ABCA1表达的影响

摘要 目的观察高糖、AGE及罗格列酮对高压力培养下THP-1源泡沫细胞ABCA1蛋白表达的影响,探讨ABCA1在高血压和糖尿病动脉粥样硬化中的作用机制。方法根据不同处理因素将THP-1源泡沫细胞分为11组:CON组,HP组,HG组,AGE组,HG-HP组,AGE-HP组,HP-R组,HG-R组,AGE-R组,HG-HR组,AGE-HR组。用Westernblot法测定其ABCA1蛋白表达。结果高糖、AGE、高压力及高压与高糖、AGE联合作用明显减弱ABCA1表达(P<0.01),罗格列酮能减弱或取消这种作用。结论THP-1源泡沫细胞ABCA1表达下调可能是高血压及糖尿病动脉粥样硬化的原因之一,其机制可能与PPARγ有关。 Objective It is to observe the influence of high glucose, AGE and Rosiglitazone on the expression of ABCA1 albumen in THP- 1 source foam cell under high pressure, in order to discuss the action mechanism of ABCA1 in hypertension and diabetic atherosclerosis. Methods THP- 1 source foam cells were divided into eleven groups including CON group, HP group, HG group, AGE group, HG- HP group, AGE - HP group, HP - R group, HG - R group, AGE - R group, HG - HR group and AGE - HR group according to different preconditioning. The expression of ABCA1 albumen was determined with Westernblot method. Results High glucose, AGE, high pressure, high pressure combined with high glucose and AGE could weaken the expression of ABCA1 (P 〈 0.01 ). Rosiglitazone could weaken or cancel the action. Conclusion The expression lowering of ABCA1 in THP - 1 source foam cell maybe one of the causes of hypertension and diabetic atherosclerosis. The mechanism maybe concerned with PPARγ.
