摘要
目的用99mTc标记抗人肺癌单克隆抗体3D3。方法以SnCl2作为还原剂直接法标记。结果采用20μgSnCl2进行标记反应,所得标记抗体标记率高(>98%),稳定性好(24h放化纯度>90%),99mTc-胶体含量<5%。体外免疫荧光分析及放射免疫结合分析提示标记抗体基本保持原抗体的免疫活性,对人肺癌组织有较高的特异性。结论该法试剂简单,快速稳定,还原型抗体可制成药盒形式,有利于临床应用。
Objective\ To label the antihuman lung cancer monoclonal antibody 3D 3 with 99m Tc.\ Methods\ It was directly labeled using SnCl 2 as a reduction agent.\ Results\ A radiolabeling yield obtained was more than 98% and 99m Tccolloid less than 5% when using 20 μg SnCl 2·2H 2O.\ In vitro studies demonstrated that the radiolabeled antibody 3D 3 was stable(24 h radiochemical purity>90%) and retained its specific binding to the human lung cancer tissue in immunofluorescence assay or radioimmunoassay.\ Conclusion\ This direct method is simple,rapid and stable.\ Reductive antibody 3D 3 can be made in a kit.\ It takes advantage of clinical application.
