摘要
本文以大肠杆菌为出发菌株,对天冬氨酸转氨酶产酶条件进行了研究。利用甲酚红为指示剂,对出发菌进行初筛,摇瓶复筛选最终得到产酶最高菌株XJ。对菌株XJ进行发酵条件的优化,得到摇瓶发酵最佳培养条件:培养基pH7.5,装液量50mL/250mL,接种量10%,培养温度38℃,摇床转速180r/min。菌株XJ在优化条件下的产酶活力(79.10mg/mL.h)比初始条件下(56.35mg/mL.h)提高了40.37%。
The enzyme-produclng conditions of Aspantate Aminotransferase by fermentation using E.coli as parent strain has been studied in this dissertation. The strain XJ was obtained by using cresol red as indicator in plate-screening and in flask-screening. The optimal fermentation conditions of Aspantate Aminotransferase for XJ were as pHT.5, 38℃,180rpm, seed volume 10%,50ml medium in 250ml flask. The activity of Aspantate Aminotransferase can reach 79.10 mg/mL.h under optimal condition ,which increasd by 40.37% than that of the parent strain(56.35mg/mL.h).
