摘要
目的探讨Ghrelin对大鼠急性胰腺炎的保护作用。方法将54只Wister大鼠随机分为假手术组、急性胰腺炎组、Ghrelin治疗组。应用牛磺胆酸钠经胆胰管逆行注射建立急性重症胰腺炎模型,再应用Ghrelin进行治疗,观察各组大鼠血清淀粉酶、TNF-α、IL-6、肠转运系数、腹腔积液及胰腺的镜下病理组织学变化。结果应用Ghrelin组中6h亚组大鼠腹腔积液较胰腺炎组减少,胰腺病理改变减轻,血清淀粉酶、TNF-α、IL-6等均较胰腺炎组明显降低,但肠转运系数无明显变化;12h亚组大鼠胰腺病理改变减轻,血清淀粉酶、TNF-α、IL-6等均较胰腺炎组明显降低,但胰腺含水量、腹腔积液、肠转运系数无明显变化;24h亚组与重症胰腺炎组比较,胰腺含水量、腹腔积液、肠转运系数、胰腺病理、血清淀粉酶变化并不明显(P>0.05)。结论Ghrelin可能对大鼠急性胰腺炎具有治疗作用,并且治疗作用可能会随Ghrelin水平的降低逐渐消退。
Objective To investigate the protective effect of Ghrelin administration on the development of acute pancreatiffs in rats. Methods 54 Wister rats were randomly divided into 3 groups: sham operation group, AP model group, Ghrelin treatment group. A modified traditional intrapancreatobiliary duet injection method was adopted using sodium taurocholate and microsyringe to make AP models. The rats were sacrificed 6h, 12h, 24h after sodium taurocholate injections, followed by the other treatments: detection of the level of serum amylase, TNF- α and IL- 6; evaluation of pathological changes of pancreas and water content of pancreas; calculation of the transit index. Results After treatment with Ghrelin for 6h, the development of pancreatitis was attenuated and the inflammatory infiltration of pancreatic tissue reduced. Also, serum amylase, TNF-α and IL-6, water content of pancreas were reclined. However, the intestinal transit index had no significant difference between the two groups. After treatment with Ghrelin 12h, the development of pancreatitis attenuated and inflammatory infiltration of pancreatic tissue reduced. In addition, serum amylase, TNF-α and IL-6 were reclined. However, there had no significant difference in water content of pancreas and transit index. After treatment with Ghrelin 24h, TNF-α and IL- 6 reduced. However, the development of pancreatitis could not attenuate, neither did the water content of pancreas, inflammatory infiltration of pancreatic tissue, serum amylase, transit index changed. Conclusions ①Administration of Ghrelin probably attenuates pancreatic damage in sodium taurocholate-induced pancreatitis; ②Protective effect of Ghrelin administration probably weakens the following concentration of Ghrelin.
