摘要
目的:研究附子提取物抗移植性肝癌H22的作用机制,并探讨与环磷酰胺是否具有增效协同作用。方法:建立移植性肝癌H22模型,随机分组,腹腔注射给药,观察各组抑瘤效应,检测TNF-α、NF-κB及Caspase-3表达。结果:附子提取物能显著抑制肝癌移植瘤的生长(P<0.05),与环磷酰胺合用可显著抑制移植瘤生长(P<0.01),有协同作用(q>1);附子提取物可显著促进移植瘤TNF-α和Caspase-3表达,抑制NF-κB表达(P<0.05),合用环磷酰胺可极显著促进TNF-α和Caspase-3表达、抑制NF-κB表达(P<0.01)。结论:附子提取物具有确切的抑瘤作用,其效应机制可能与活化细胞凋亡信号传导通路,诱导肿瘤细胞凋亡有关,并与环磷酰胺有协同增效作用。
Objective:to study the functional mechanism of Radix Aconiti Praeparata Extract against transplanted liver cncer H22,and probe into its synergistic interactions with cyclophosphamide.methods:transplanted liver cancer H22 model were established and randomly diveided into different groups.After the intraperitoneal injection,observe the tumor-inhibiting effect of each group,detect the expressions of TNF-α,NF-κB and Caspase-3,Results:The Radix Aciniti Praeparata Extract can obviously inhibit the growth of transplanted tumor(P〈0.01),with synergistic effect(q〉1),Radix Aconiti Praeparata Extract can obviously improve the expressions of transplanted tumor TNF-α and caspase-3,inhibit the expression of NF-κB (P〈0.05),its application together with cyclophospharmide can significantly improve the expressions of TNF-α and caspase-3,inhibit the expression of NF-κB(P〈0.01),Conclusion:Radix Aconiti Praeparata Extract has exact tumor-inhibiting effect,its effect machanism may be related to the transmission passway of the activated cell apoptosis signal and tumor cell apopto sis induction,and have synergistic effect with eyclophosphamide.
