强直性脊椎炎与HLA-B27等位基因研究

STUDY ON ANKYLOSING SPONDYLITIS AND HLAB27 ALLELES

为探索强直性脊椎炎与ＨＬＡ－Ｂ２７关联机理，建立非同位素地高辛标记的ＰＣＲ／ＳＳＯ方法对中国上海地区６８例ＨＬＡ－Ｂ２７阳性的强直性脊椎炎病人和７例ＨＬＡ－Ｂ２７阳性正常对照进行了ＨＬＡ－Ｂ２７等位基因的检测，结果显示在病人和正常对照组中Ｂ＊２７０４都是最常见的等位基因，各占７６．８％和７１．４％，Ｂ＊２７０５亦是常见的等位基因，分别占２０．３％和２８．６％，两组间比较差异无显著性；在病人组还检出Ｂ＊２７０６和Ｂ＊２７０７各１例，提示此两等位基因可能也与ＡＳ关联。此结果为研究强直性脊椎炎与ＨＬＡ－Ｂ２７关联的机制提供了有用的资料。

To explore the mechanism of association between ankylosing spondylitis and HLAB27,had investigated the distribution of HLAB27 alleles in 68 cases of B27 positive AS patients and 7 cases of B27 positive healthy controls using nonradioactive digoxigenin labelled PCR/SSO method.The result showed that B*2704 is the most common allele in both patient and control groups(76.8% and 71.4% respectively).B*2705 is also a common allele with allele frequences 20.3% and 28.6% seperately.Neither of the two alleles is significantly different between patients and controls.Interestingly,had detected case of one B*2706 allele and one B*2707 allele in patient group,which suggested these two alleles may be associated with AS.
