固本生血丸对人骨髓造血祖细胞体外增殖的影响

THE EFFECTS OF “GU BEN SHENG XUE WAN”ON THE PROLIFERTIN OF HEMATOPOIETIC PROGENITOR CELLS FROM HUMAN MARROW

应用造血祖细胞体外培养法研究了中药复方制剂——固本生血丸对人骨髓造血祖细胞体外集落形成的影响。结果表明：该制剂在０．０１～１０μｇ／ｍｌ浓度范围内与细胞因子（ＧＭ－ＣＳＦ或ＥＰＯ）协同促进正常骨髓和再障骨髓ＣＦＵ－ＧＭ、ＨＣＦＵ－ＧＭ，ＣＦＵ－Ｅ和ＢＦＵ－Ｅ的集落形成；作用高峰在１０μｇ／ｍｌ浓度上；当超过该浓度时，集落数量增加不明显（Ｐ＞０．０５），而单独应用该制剂时无集落形成。

The effects of “Gu Ben Sheng Xue Wan” on proliferation of hematopoietic progenitor cells from human marrow were studied in a vitro semisolid culture.The finding showed that “Gu Ben Sheng Xue Wan” had synergistically stimulated in vitro the formation of colonies from CFUGM(ColonyForming UnitGranulocyteMacrophage),HCFUGM(Huge CFUGM),CFUE(ColonyForming UnitErythrocyte) and BFUE(Erythrocytic Burst ColonyForming Unit) from normal human and AA(Aplastic Anemia)marrow with GMCSF(Colony Stimulating FactorGranulocyteMacrophage)and EPO(Erythopoietin) in an appropriate range of the concentration(0.01～10 μg/ml).The largest increase of the numbers of the colonies was observed when its concentration was at 10 μg/ml.However,no obvious effects on formation of colonies were obsvered when its concentration was in excess of 10 μg/ml.It had not stimulated the formation of the colonies from CFUGM,HCFUGM,CFUE and BFUE alone.
