摘要
应用造血祖细胞体外培养法研究了中药复方制剂——固本生血丸对人骨髓造血祖细胞体外集落形成的影响。结果表明:该制剂在0.01~10μg/ml浓度范围内与细胞因子(GM-CSF或EPO)协同促进正常骨髓和再障骨髓CFU-GM、HCFU-GM,CFU-E和BFU-E的集落形成;作用高峰在10μg/ml浓度上;当超过该浓度时,集落数量增加不明显(P>0.05),而单独应用该制剂时无集落形成。
The effects of “Gu Ben Sheng Xue Wan” on proliferation of hematopoietic progenitor cells from human marrow were studied in a vitro semisolid culture.The finding showed that “Gu Ben Sheng Xue Wan” had synergistically stimulated in vitro the formation of colonies from CFUGM(ColonyForming UnitGranulocyteMacrophage),HCFUGM(Huge CFUGM),CFUE(ColonyForming UnitErythrocyte) and BFUE(Erythrocytic Burst ColonyForming Unit) from normal human and AA(Aplastic Anemia)marrow with GMCSF(Colony Stimulating FactorGranulocyteMacrophage)and EPO(Erythopoietin) in an appropriate range of the concentration(0.01~10 μg/ml).The largest increase of the numbers of the colonies was observed when its concentration was at 10 μg/ml.However,no obvious effects on formation of colonies were obsvered when its concentration was in excess of 10 μg/ml.It had not stimulated the formation of the colonies from CFUGM,HCFUGM,CFUE and BFUE alone.
