沙美特罗平喘作用的研究

Antiasthmatic Action of Salmeterol

沙美特罗雾化吸入给药，在剂量为１２５μｇ／ｋｇ、２５０μｇ／ｋｇ、５００μｇ／ｋｇ时，可明显延长由乙酰胆碱和组胺引起的豚鼠引喘潜伏期，且抽搐的豚鼠数目也明显减少。沙美特罗的平喘作用具有剂量依赖性。离体实验证明，沙美特罗在浓度为３×１０８ｍｏｌ／Ｌ及３×１０６ｍｏｌ／Ｌ时，均可使组胺（浓度为３×１０４ｍｏｌ／Ｌ）引起的气管收缩幅度下降，抑制率分别为（２５９６±３２５）％、（６３９３±７２７）％。

Inhaled salmeterol at doses of 125μg/kg,250μg/kg,500μg/kg could obviously prolong the latent periods of asthma and reduce the numbers of convulsive animals induced by acetycholine and histamine in guinea pig.The antiasthmatic action was dose dependent.The experiments in vitro showed that salmeterol at concentrations of 3×10 8 mol/L and 3×10 6 mol/L could inhibit the action of bronchocontraction induced by histamine,the inhibition rates were (25.96±3.25)% and (63.93±7 27)% respectively.
