人卵透明带3蛋白的二级结构和B细胞抗原表位预测 被引量:3

摘要 目的:预测并综合分析人卵透明带3(human zona pellucida 3,hZP3)蛋白的二级结构及其B细胞抗原表位。方法:以hZP3蛋白的基因序列为材料,采用Garnier-Robson法、Chou-Fasman法和Karplus-Schulz法预测其结构蛋白的二级结构及蛋白骨架区的柔韧性,采用Kyte-Doolittle法对蛋白的亲水性进行分析,Emini法预测蛋白的表面可能性,Jameson-Wolf法预测蛋白的抗原指数。结果:hZP3蛋白含有较多的β折叠和转角结构。该蛋白第27~31、36~46、114~133、140~151、237~240和324~329区段可能是α-螺旋中心;hZP3蛋白分子第52~54、60~67、74~78、100~111、158~169、181~185、190~193、210~218、226~229、311~317、335~349和353~360区段可能是β-折叠中心。hZP3蛋白具有多处抗原指数较高的区段,其中近中部区段和近羧基端的第92~95、132~137、171~178、239~245、252~255、279~286、292~305和319~324区段含有潜在的B细胞优势抗原表位。结论:以蛋白质的二级结构预测作为辅助手段,用抗原指数、亲水性参数和表面可能性参数预测hZP3蛋白的B细胞抗原表位,为实验确定hZP3蛋白的B细胞抗原表位并以此进行免疫避孕研究奠定基础。 Objective: To predict and analyze the secondary structure and B cell epitopes of zona pellucida glycoprotein 3 in Homo Sapiens (hZP3). Methods: The secondary structure and flexible regions of hZP3 protein were predicted by the methods of Garnier- Robson, Chou Fasman and Karplus-Schulz based on hZP3 gene sequence. Moreover, hydrophilieity plot, surface probability plot and antigenic index of hZP3 protein were predicted by the methods of Kyte-Doolittle, Emini and Jameson-Wolf, respectively. Results: hZP3 protein contained more t; sheet and turn regions. There are several centers of α helix in the No. 27~ 31, 36~46, 114~133, 1404151, 237~240 and 324~329 of hZP3 protein. There are more centers of sheet in the No. 52~54, 60~67, 74~78, 100~111, 158~169, 181~185, 190~193, 210~218, 226~ 229, 311~317, 335~349 and 353~360 ofhZP3 protein. Moreover, theB cellepitopes possibly localized in or nearbyhZP3 protein's central and C terminal region of No. 92~95, 132~137, 171~-178, 239~245, 252~255, 2794286, 292~305 and319~324. Conclusion: These results are helpful for identification of B cell epitopes for hZP3 protein and the research of immunocontraception based on these epitopes using experimental methods.
