摘要
目的:研究脊髓腹侧压迫损伤后脊髓内部构筑变化及c-Fos蛋白的表达情况。方法:取健康新西兰大白兔24只,建立脊髓腹侧压迫损伤模型,分为压迫组和对照组,依次于造模手术后0.5、1、2、4h取材。取脊髓作连续切片后,采用HE及免疫组化方法,观察压迫后不同时间脊髓内部的病理变化与脊髓前角c-FOS阳性神经元的表达情况。结果:(1)造模组脊髓内部构筑出现不同程度的位移、偏转和变形,尤以灰质Ⅷ、Ⅸ板层明显,病变程度随压迫时间的增加而加重。(2)脊髓腹侧损伤后c-Fos阳性神经元细胞在脊髓前角、中间带和中央管周围的大多角形细胞以及后角的小圆细胞均有表达,其表达尤以前角Ⅷ、Ⅸ板层显著。压迫组c-Fos阳性神经元细胞数量与对照组之间差异有统计学意义(P<0.05)。结论:脊髓腹侧压迫后,脊髓的外部形态和内部构筑均发生了变化、位移和不同程度的病理变化,尤以Ⅷ、Ⅸ板层比较明显,Ⅶ板层变化次之。c-Fos阳性神经元细胞在脊髓前角Ⅷ、Ⅸ板层的大多角形细胞呈高表达,提示c-Fos蛋白的表达可能与脊髓构筑变化有关。
Objective:To research the changes of internal structure of spinal cord and c-Fos protein expression after abdominal-side compressive trauma of spinal cord.Methods:Twenty four healthy New Zealand rabbits were selected to establish the traumatic model of abdominal side compression on spinal cord and classified into compression group and reference group respectively.Sampling was taken 0.5 hour,1 hour,2 hours and 4 hours after operation.Spinal cord was sampled and sliced up continuously with HE and IHC method adopted to observe the pathological changes of spinal cord in different time after compression,as well as the expression of c-Fos positive neurons in anterior horn of spinal cord.Results:(1)In experimental group,the internal construction of spinal cord showed various changes in different degrees,such as backward shifting,lateral deviation and deformity,especially obvious in anterior horn(Ⅷ,Ⅸ plate).The traumatic conditions were exacerbated when duration of compression was prolonged.(2)After abdominal-side trauma of spinal cord,c-Fos positive neurons were expressive as following patterns especially obviously in anterior horn(Ⅷ,Ⅸ plate):the big multi-angular cells surrounding anterior horn of spinal cord,central belt and central canal,the small round cells in posterior horn.The comparison of c-Fos positive neurons numbers was statistically significant(P〈0.05)between compression group and reference group.Conclusion:After abdominal-side compression of spinal cord,changes,including shifting and various pathological changes,took place both in external morphology and internal construction of spinal cord,more obvious in Ⅷ,Ⅸ plate and less obvious in Ⅶ plate.c-Fos positive neurons cells were highly expressive in the big multi-angular cells surrounding anterior horn,especially obvious in Ⅷ,Ⅸ plate,indicating the possible correlation between the construction changes of spinal cord and c-Fos protein expression.
